I fell in love with podcasts again recently, mostly thanks to Marc Maron's WTF Podcast. A comic himself for many years (considered the original 'alt comic'), Mark's honest and deep interviews with comics are amazing listens. Guests have included Louis CK, Judd Apatow, Eugene Mirman, Dane Cook, Adam McKay, Ira Glass, and a 2 part Carlos Mencia podcast which is essential if you follow comedy and stories of comics stealing other people's work.

Seriously, it is worth subscribing to. Some people describe it as "Charlie Rose but with comedians" but I think it's much deeper than that.
